Simultaneous Determination of Carotenoids and Chlorophylls by the HPLC-UV-VIS Method in Soybean Seeds

Soybean contains nutritional bioactive compounds, including carotenoids associated with human health benefits. Carotenoids are applicable in pharmaceuticals/nutreceuticals, cosmetic, and mainly food industries. However, an efficient accurate method for carotenoid chlorophyll detection quantification has not yet been developed validated soybean seeds. The need a rapid reliable analysis become increasingly important. Thus, this study was initiated to develop validate simple, rapid, selective reversed-phase high-performance liquid chromatographic (RP-HPLC) the simultaneous determination of lutein, zeaxanthin, ?-carotene, ?–carotene, ?–cryptoxanthin, chlorophyll–a –b flour sample (100.00 mg) extracted using ethanol-acetone (1:1) solvents at volume 1.50 mL. Interestingly, effective separation technique achieved mobile phases methyl tert-butyl ether, methanol containing 10 mM ammonium acetate, water delivered 0.90 mL min?1 flow rate through C30YMC Carotenoid (250 × 4.6 mm I.D., S-5 µm) column coupled UV-VIS detector set 450 nm. response linear from 0.05–30.00 ?g mL?1 coefficient (R2) 0.9993–0.9999. sensitive limit (LOD) 0.0051–0.0300 0.0155–0.0909 (LOQ). recovery values were 83.12–106.58%, repeatability precision ranged 1.25–4.20% 0.15–0.81% system, respectively. showed adequate relative standard deviation smaller than 3.00%. This also found be profiling chlorophylls other legumes. In summary, successfully implemented qualitative quantitative major legume seeds, which beneficial industry quality breeding programs meet nutrition demands globally.
